Anil-Babesia rodhaini Monoclonal Antibodies: Effect against Babesia rodhaini and Cross-reactivity with Human Starin of Babesia microti OAK

Anti-Babesia rodhaini monoclonal antibodies (mAb), namely: 1-E7, 2-H2 and 3-B8, significantly suppressed the development of high parasitemia in BALB/c mice infected with B. rodhaini and all mAb-treated mice survived the infection. While, only monoclonal antibody 3-B8 showed some inhibitory effect against Babesia microti, with mice showing high parasitemia of 18.04 ± 2.69 %, at 9 days post-exposure. Westernblot analysis of B. rodhaini and B. microti parasite extract reacted with anti-B. rodhaini monoclonal antibodies showed cross-reactive bands of molecular weights 62 and 55 kilodaltons. Artesunate, a potential drug for treatment of Babesia infection OAK

The effect of artesunate, a water-soluble artemisinin derivative, against Babesia species, such as Babesia bovis, Babesia gibsoni, and Babesia microti was studied. Cultures of B. bovis and B. gibsoni were treated with 0.26 μM, 2.6 μM, 26 μM, and 260 μM artesunate, and the growth-inhibitory effect was shown in over 2.6 μM artesunate in day 4 and day 3 post-subculture for B. bovis and B. gibsoni, respectively, in dose-dependent manner. In vivo experiment for B. microti, strong inhibition effects were observed in mouse groups treated with over 1.0 mg/kg body weight of artesunate on day 9 and 10 post-infection. These results suggest that artesunate could be a potential drug for Babesia infection.

Changes of splenic interleukin-12p70 concentration in mice infected with Babesia microti and Babesia rodhaini OAK

Interleukin-12 (IL-12) is one of the most important factors for the host’s protective immune response against protozoan infection. To evaluate the effect of bioactive form of IL-12(IL-12p70), expression of IFN-AmRNA and IL-4 mRNA in splenic CD4 positive T cells, anti-parasite delayed type hypersensitivity (DTH), and serum and splenic IL-12p70 concentration were examined in Babesia microti and B. rodhaini infected mice. The expression of IFN-AmRNA as a marker of Th1 cell differentiation was detected in splenic CD4 positive T cells in B. microti infected mice. Activation of DTH response as a marker of cell-mediated immune response was also observed from 2 days after the infection with B. microti, while no response was detected in B. rodhaini infected mice....

Oligomannose-coated liposome (OML)-based vaccines have been reported to induce Th1-based immunity against entrapped antigens in immunized animals and to show protective effects against several protozoal diseases. In the present study, we produced a recombinant Babesia rodhaini ribosomal phosphoprotein P0 (rBrP0) that showed immunological cross reactivity with another rodent Babesia parasite, B. microti. We evaluated the efficacy of vaccination with OML-entrapped rBrP0 on B. rodhaini and B. microti infections in mice. Babesia microti is a rodent tick-borne blood parasite and the major causative agent of emerging human babesiosis. Here, we identified a candidate of common antigenic protein BmP41 of B. microti by serological screening of cDNA library of human-pathogenic Gray strain with antisera against rodent Munich strain. Immunofluorescent antibody test using mouse anti-recombinant BmP41 (rBmP41) serum revealed that native BmP41 was expressed in each of the developmental stages of B. microti merozoites. An enzyme-linked immunosorbent assay (ELISA) using rBmP41 detected specific antibodies in sera from hamsters infected with B. microti Gray strain and mice infected with B. microti Munich strain. PCR detection of Babesia microti and Babesia divergens in ticks OAK

We investigated the presence of Babesia microti and B. divergens in Ixodes ricinus ticks collected in a Lyme borreliosis endemic area of Northern Poland. We used as the PCR target the fragment from a gene encoding the nuclear small-subunit ribosomal RNA (SS-rDNA). We have made a thorough analysis of the quantitative and rate per cent occurrence Ixodes ricinus from two areas of forest in province of Szczecin, known as highly recreative and frequented of many people resort. A total of 533 specimens collected in 13.3% was detected DNA of B. microti and in 3.0% of B. divergens.
